Some these were reported cause leaf symptoms yield reduction. In 2009 2013, surveys performed on orchards located northern presence Betaflexiviridae using generic specific molecular assays (Foissac et al. 2005). Sanger a reverse transcription polymerase chain reaction (RT-PCR) product originated from sample collected symptomless (cv. Ferrovia) revealed virus sequence sharing 75% nucleotide (nt) similarity with highly conserved portion betaflexivirus gene. Total RNA, extracted TRIzol reagent (Invitrogen, Carlsbad, CA) leaves sample, was subjected HTS analysis an Illumina Hi-seq 4000 platform (Novogene, Tiangen, China). The run, after quality control, yielded 26,002,319 150-nt-long paired-end reads. De novo assembly reads Geneious Prime 2019.2.3 Velvet subsequent BLAST (BLASTn/x) 82 contigs revealed, apart several known (cherry A, Prunus F, little 1), newly described Robigovirus species. More specifically, results that eight de ranging 277 2,287 nt long similar recently (CVTR) (Çağlayan 2019). nearly complete genome consisting 8,362 (99.5%) Greek CVTR isolate (CVTR-C18GR, accession no. MT043307) reconstructed iterative mapping contigs, 4,690 mapped total RT-PCR amplicons fill gaps. Similarity CVTR-C18GR full sequences GenBank CVTR-BUR12 (MK600387) CVTRAZQ (MH177869) showed 90.47 90.60% identities, respectively. Phylogenetic analyses confirmed clustering C18GR other isolates. To investigate Greece, primers F2Rob (5′-GTCAGAGAGAGGTATCTATGTC-3′) RT1Rob (5′-CAGGCTGTTCATAACCT-3′), which amplify 577 replicase, designed. Sixty-six samples trees showing no virus-like various screened CVTR. Eight them found be infected new virus. C11GR amplicon fields (96% C18GR, MT043308). Sequence between isolates also high (97% nt) partial TGB2, TGB3, CP genomic regions amplified primer pair 7006 F (5′-GAAAAGTGATTATTCAGCRCCAGT-3′) 7708 R (5′-CTTTCACCCACTCATCACCTATCTCC-3′) (accession MT461407). This is first report thus extending information its geographical distribution. Robigoviruses variable diseases sour cherries, reason they are included European Commission Directive concerning official inspections (2019/2072/EU). Sweet cherry rootstocks for the Pacific Northwest

All commercial sweet cherry trees are either budded or grafted. The part of the tree above the graft/bud union is known as the scion and the part below the graft/bud union is known as the rootstock. Sweet cherry scion cultivars have been selected over millennia for many reasons, but over the past century, breeding programs have concentrated mainly on achieving improved characteristics such as y...

Genetic Diversity in the Coat Protein Genes of Prune dwarf virus Isolates from Sweet Cherry Growing in Turkey

Sweet cherry is an important fruit crop with increasing economical value in Turkey and the world. A number of viruses cause diseases and economical losses in sweet cherry. Prune dwarf virus (PDV), is one of the most common viruses of stone fruits including sweet cherry in the world. In this study, PDV was detected from 316 of 521 sweet cherry samples collected from 142 orchards in 10 districts ...

Determination of self-incompatibility groups of sweet cherry genotypes from Turkey.

Determination of S-allele combinations of sweet cherry genotypes and cultivars has importance for both growers and breeders. We determined S-allele combinations of 40 local Turkish sweet cherry genotypes using a PCR-based method. Ten different S-alleles were detected. Although the most common S-allele was S3, as also found in Western genotypes and cultivars, there were some differences in the f...

First Complete Genome Sequence of Cherry virus A

The 5'-terminal genomic sequence of Cherry virus A (CVA) has long been unknown. We determined the first complete genome sequence of an apricot isolate of CVA (7,434 nucleotides [nt]). The 5'-untranslated region was 107 nt in length, which was 53 nt longer than those of known CVA sequences.
